Synthèses de béta-lactames mono et bicycliques par voies ionique et radicalaire

Ce travail présente deux approches pour la synthèse de méthoxy-3-mono-bactames et méthoxy-6 carbapénames à partir de produits naturels : l'acide L(+) tartrique et la D-glucosamine. Dans le premier chapitre nous étudions la réaction de décarboxylation radicalaire de l'acide tartrique en présence d'une oléfine activée, conduisant à la formation d'une liaison carbone-carbone avec un excellent contrôle de stéréochimie. Dans le second chapitre nous décrivons une synthèse originale de méthoxy-3 monobactames à partir de l'acide L(+) tartrique, basée la décarboxylation radicalaire décrite dans le premier chapitre et sur la méthylation régiosélective d'un diol vicinal. Dans la première partie du troisième chapitre nous présentons une deuxième approche pour la synthèse chirale de méthoxy-3 monobactames ; celle-ci met en jeu une cycloaddition (2+2) d’une imine dérivée de la D-glucosamine et du cinnamaldéhyde, et d’un cétène dérivé de l’acide méthoxyacétique. Dans la deuxième partie de ce chapitre nous décrivons leurs transformations en : a) carboxy-4 methoxy-3 monobactame en vue d'étudier sa décarboxylation radicalaire en présence d'une oléfine activée ; b) methoxy-6 carbapéname, en une seule étape, à partir des composés issus de la cycloaddition (2+2), par l’utilisation d’une réaction de cyclisation radicalaire intramoléculaire.
This work presents two approaches to the synthesis of 3-methoxy monobactams and 6-methoxy carbapenams starting from two natural products: L(+) tartaric acid and D glucosamine. In the first chapter, the radical decarboxylative alkylation of tartaric acid leading to the formation of a carbon-carbon bond with almost complete retention of configuration is studied. In the second chapter we present a novel synthesis of 3-methoxy monobactams starting from L(+) tartaric acid, based on the work described in the first chapter and the regioselective methylation of a vicinal diol. In the first part of the third chapter we present a second approach to the synthesis of 3-methoxy monobactams; this method involves a (2+2) cycloaddition carried out with an imine formed from D-glucosamine and cinnamaldehyde, and a ketene derived from methoxyacetic acid. In the second part of this chapter we describe their transformation to: a) 4-carboxy 3-methoxy monobactam to study its radical decarboxylative alkylation. B) 1-benzyl 6-methoxy carbapenams, obtained in one step by radical intramolecular annulation from the (2+2) cycloaddition products.
